PINK1 Deficiency Aggravates the β-amyloid-attenuated Mitophagy-lysosomal Degradation in PC12 Cells

چکیده

Background and objectivePTEN-induced putative kinase 1 (PINK1) is a mitochondrial that regulates mitophagy. PINK1-deficient mAPP mice display low LC3B levels, PINK1 overexpression enhances autophagy increases the expression level of lysosome-associated membrane protein (LAMP1). The present study evaluated whether altered could modulate β-amyloid (Aβ)-treated mitophagy in PC12 cells, simple cellular model to simulate pathological changes neurodegenerative diseases vitro.

Lysosomal NEU1 deficiency affects Amyloid Precursor Protein levels and amyloid-β secretion via deregulated lysosomal exocytosis

Alzheimer's disease (AD) belongs to a category of adult neurodegenerative conditions, which are associated with intracellular and extracellular accumulation of neurotoxic protein aggregates. Understanding how these aggregates are formed, secreted and propagated by neurons has been the subject of intensive research, but so far no preventive or curative therapy for AD is available, and clinical t...

LINGO-1 promotes lysosomal degradation of amyloid-β protein precursor

Sequential proteolytic cleavages of amyloid-β protein precursor (AβPP) by β-secretase and γ-secretase generate amyloid β (Aβ) peptides, which are thought to contribute to Alzheimer's disease (AD). Much of this processing occurs in endosomes following endocytosis of AβPP from the plasma membrane. However, this pathogenic mode of processing AβPP may occur in competition with lysosomal degradation...

Mitophagy Regulated by the PINK1-Parkin Pathway

Mitochondria play key roles in the cellular metabolism of lipids and iron as well as in cell death signaling. Mitochondrial dysregulation produces reactive oxygen species (ROS), which results in oxidative stress. Moreover, the accumulation of damaged mi‐ tochondria leads to cell death and tissue dysfunction. Mitochondrial maintenance in‐ volves mitophagy, a selective autophagy process that remo...

Mitochondrial and lysosomal biogenesis are activated following PINK1/parkin‐mediated mitophagy

Impairment of the autophagy-lysosome pathway is implicated with the changes in α-synuclein and mitochondrial dysfunction observed in Parkinson's disease (PD). Damaged mitochondria accumulate PINK1, which then recruits parkin, resulting in ubiquitination of mitochondrial proteins. These can then be bound by the autophagic proteins p62/SQSTM1 and LC3, resulting in degradation of mitochondria by m...

Mitochondrial dysfunction activates lysosomal-dependent mitophagy selectively in cancer cells

Molecules designed to target and accumulate in the mitochondria are an emerging therapeutic approach for cancer and other indications. Mitochondria-targeted redox agents (MTAs) induce mitochondrial damage and autophagy in cancer cells. However, the mechanisms for these molecules to induce mitophagy, the clearance of damaged mitochondria, are largely unknown. Using breast derived cell lines and ...
